The Serampore trio—three Baptist missionaries—founded the institution with the goals of educating students from all "castes, colours, creeds, or countries" in the arts and sciences and preparing them for ministry in India's expanding Christian community.

On February 23, 1827, in Copenhagen, Denmark, King Frederick VI of Denmark granted Serampore College its Royal Charter of Incorporation since Serampore was then a Danish colony (Charter, 1, Charter, 2, Charter, 3). The Baptist missionary Joshua Marshman visited King Frederick in August 1826, prompting the charter, which granted Serampore College the right to confer degrees in both theology and the arts. The first Council included Baptist missionaries William Carey, Joshua Marshman, and John Clark Marshman (Joshua's son). The Trio published a prospectus at the time of its founding that suggested "A College for the teaching of Asiatic Christian and other Youth in Eastern Literature and European Science."
