DIPSAR was established in Delhi's Kashmiri Gate in 1964 as the Department of Pharmacy. It was transformed into the College of Pharmacy and relocated to the Pusa Institute site in 1972. It finally relocated to its current site in Pushp Vihar in 1979. It became affiliated with University of Delhi in 2004 and was given the name Delhi Institute of Pharmaceutical Sciences & Research.

The Delhi Government passed the Delhi Pharmaceutical Science and Research University in 2008. The institute didn't officially open its doors until 2015, with DIPSAR serving as both of its member colleges and both of them occupying the same space.
